Trace Metals as Drivers of Microbial Success and Laboratory Failure

Abstract: Trace metals play a crucial role in the metabolism of all living organisms and many metal-containing enzymes contribute to key physiological and ecological processes. Despite their importance, the precise impact of trace metal availability on microbial functional diversity and metabolic shifts remains poorly understood. Using Escherichia coli, we show that trace metal availability dictates electron acceptor utilization, even under energetically favorable conditions. Our results reveal that trace metals act as gatekeepers, as their availability is crucial for unlocking the full metabolic potential and growth success of microbes. Concurrently, we identify a critical methodological vulnerability introduced by hidden contamination from common laboratory equipment, consumables, and chemical reagents. Our results show that a rigorous monitoring of trace element abundance is paramount to prevent laboratory failure and ensure reproducibility
