Sonnante Giuseppe has got his high school diploma as a Chemical Technician during the school year 1973-74. In the period 01/12/1976-31/03/1977 he has worked, as a technician, at the University of Bari, Institute of Agricultural Industry. His activity was devoted to the study of oxidative processes in olive oil.

In 1977 he was hired as a technician at the CNR, Water Research Institute (IRSA) of Bari. He worked there on several topics, until February 2003, and particularly on: separation processes by means of reverse osmosis; removing of heavy metals from water; removing of phosphorus by means of biological treatments; study on take up of organic pollutants on heterogeneous solid phases; treatment of chemical oxidation for potable water; set up of the best conditions for the chromatographic detection of pesticide residues; analytic methods for the detection of aromatic compounds in sea sediments.

Moreover, at CNR-IRSA, he was in charge of the following tasks:

  • Teacher, as an expert, of “Heavy metals removing” at the high school ITIS “Modesto Panetti”, in Bari;
  • tasks attributed by the Director to perform chemical analyses on sea sediments within the frame of a collaboration between ENEL and IRSA, in 1993 and 1995;
  • teacher, as an expert, of “Quality control in Agro-food, environment” at the high school IPSIA “N. Chiarulli”, Acquaviva delle Fonti, Bari, school year 1994-95.On 05/03/2003 he moved to CNR-Institute of Plant Genetics (IGV), from 14/10/2013 Institute of Biosciences and BioResources (IBBR), where he is still working at present, on the following topics:
  • handling of seeds withdrawn from Cynara heads, their conservation, germination, growing in greenhouse and in the field;
  • DNA extraction and purification from plant samples from various species (artichoke, wheat, legumes); agarose gel electrophoresis and DNA visualization by means of gel imaging systems; DNA quantification by means of Nanodrop ND 1000 spectrophotometer; PCR amplification; purification of amplificates and their cloning in plasmid vectors; preparation of PCR fragments and clones for sequencing (sequencing amplification, purification by alcohol precipitation); sequencing using Beckman CEQ 8800 Sanger sequencer; microsatellite (SSR) and AFLP marker amplification and detection on automated sequencer;
  • data analysis: use of software GenomeLab CEQ 8800 Genetic Analysis System and Microsoft Office.At CNR-IBBR, besides technical-scientific activities, Mr. G. Sonnante has been in charge of the: organization and maintenance of samples in the IGV DNA bank; management and maintenance of the sequencer Beckman CEQ 8800; use and management of the gel image system GEL LOGIC 100 KODAK; management of the laboratory, with the task of stocking reagents and consumables; purchase order enquiry and preparation; greenhouse organization.
